Genome-wide assessment of DNA methylation fidelity identifies representative ovarian cancer cell line models

Epithelial ovarian cancer comprises multiple histotypes, yet cell lines are often labelled simply “ovarian carcinoma” or “serous”. This study compares Infinium 450K and EPIC methylation profiles of 60 ovarian cancer cell lines with 108 ovarian tumours by correlation and effect-size analysis across genetic and epigenetic domains. Methylation is highly conserved between cell lines and tumours at CpG-dense regulatory regions, and specific cell lines emerge as representative models of high-grade serous, clear cell, endometrioid, mucinous and mixed histotypes.
